An efficient method for counting DAPI-stained cells using Fiji

چکیده
Counting cells is a crucial procedure in evaluating the success of a treatment. A fast yet exact way of doing this for DAPI-stained cells with the ImageJ-derivate ‘Fiji’ is described in this paper, written as a step-by-step tutorial with screenshots.

Rapid automated cell quantification on HIV microfluidic devices.
Lab-chip device analysis often requires high throughput quantification of fluorescent cell images, obtained under different conditions of fluorescent intensity, illumination, focal depth, and optical magnification. Many laboratories still use manual counting--a tedious, expensive process prone to inter-observer variability. The manual counting process can be automated for fast and precise data ...
متن کاملOptimal staining and sample storage time for direct microscopic enumeration of total and active bacteria in soil with two fluorescent dyes.
Direct counting techniques, first developed for aquatic samples, can be used to enumerate bacteria in soil and groundwater sediments. Two fluorescent dyes, 5-cyano-2,3-ditolyl tetrazolium chloride (CTC) for actively respiring bacteria and 4(prm1),6-diamidino-2-phenylindole (DAPI) for total bacteria, were tested for their usefulness in epifluorescent direct bacterial enumeration in soil. Both dy...
متن کاملEffect of fluorochromes on bacterial surface properties and interaction with granular media.
Simple, efficient, and safe tagging methods are desired in short-term microbial transport studies such as in the study of filtration systems for water and wastewater treatment. Suitability of selected fluorochromes as bacterial tagging agents in transport studies was evaluated on the basis of stability of stained cells and the effect of staining on bacterial surface characteristics and interact...
متن کاملEarly detection of Chlamydia trachomatis using fluorescent, DNA binding dyes.
HeLa 229 cells were infected with genital tract strains of Chlamydia trachomatis. After incubation for varying times the infected cells were fixed and stained with the fluorescent DNA binding dyes Hoechst 33258 or DAPI for comparison with conventional Giemsa stain. Fluorochrome-treated preparations were examined by incident ultraviolet fluorescence microscopy and the Giemsa-stained preparations...
